Step 1: Dissolve 2-amino-5-bromo-1,3-thiazole (1.0 g, 4.8 mmol) and 2-bromo-2′-chloroacetophenone (1.3 g, 4.8 mmol) in methanol (20 mL) and add hydrochloric acid (1 M) dropwise until the pH reaches 3., Step 2: Stir the reaction mixture at room temperature for 24 hours., Step 3: Add sodium hydroxide (1 M) dropwise until the pH reaches 10., Step 4: Extract the product with ethyl acetate (3 x 20 mL) and dry the organic layer over anhydrous sodium sulfate., Step 5: Concentrate the organic layer under reduced pressure to obtain a yellow solid., Step 6: Dissolve the yellow solid in methanol (10 mL) and add sodium borohydride (0.5 g, 13.2 mmol) slowly., Step 7: Stir the reaction mixture at room temperature for 24 hours., Step 8: Quench the reaction with water (10 mL) and extract the product with ethyl acetate (3 x 20 mL)., Step 9: Dry the organic layer over anhydrous sodium sulfate and concentrate under reduced pressure to obtain a white solid., Step 10: Recrystallize the white solid from methanol to obtain pure Bromazolam.
